What is GW0742 ?

GW0742 is a PPARδ/β agonist very similar in molecular structure to GW501516 (Cardarine) — with the exception of one atom.

Aside from PPARδ/β receptor activation, it has been shown to weakly affect other receptors, with in silico studies indicating thyroid activity.

It is currently being researched as an anti-diabetic drug in addition to its anti-inflammatory effects.

For bodybuilders, GW501516 (Cardarine) is often used to accelerate fat loss as it helps convert and mobilize stored fat into energy.

Another added benefit is to increase oxidative capacity in muscles, which, along with angioneogenesis, would contribute to improved endurance in athletes.

In my personal experience, Cardarine has aided to increase my cardiovascular endurance in the gym and on the treadmill, while studies support Cardarine’s efficiency in improve cardiovascular output.

Which is Better: GW0742 vs GW501516 Cardarine

Some have touted GW0742 as being Cardarine 2.0. Similar to how SR9011 was developed in the wake of Stenabolic’s poor bioavailability and frequent sublingual redosing, GW0742 has boasted significant potential — although human studies would need to be carried out to ascertain these benefits.

As discussed in a previous article, Cardarine has a number of benefits, which have been confirmed in scientific studies:

  • Increased endurance
  • Fat burning
  • Healing
  • Increased blood flow

Research has shown that the PPARδ receptor which Cardarine targets is responsible for metabolic maintenance, in particular with regard to skeletal programming, would healing and mitochondrial respiration. Genes associated with contractile proteins, mitochondrial creation and lipid oxidation are upregulated, with benefits for endurance and fat loss.

However, other studies demonstrated, at inordinately high doses, that Cardarine may have carcinogenic properties.

Drawbacks of GW0742

Now, this is a massive drawback: some studies have shown that heart enlargement in mice can occur up to 24 hours after ingestion, through the activation of calcineurin, making GW0742 a potentially dangerous compound to run.

Moreover, due to the chemical structure of GW0742 being very similar to GW501516, there is a cancer risk associated with it. However, it has to be clarified that mice that exhibited tumors were given enormous doses during a prolonged exposure to Cardarine which no person would ever run.

Animal testing in 2007 showed that the Cardarine dramatically improved the physical performance of the animals it was given to, which led to the immediate growth of a black market for the drug. At or around the same time, animal testing was abandoned by GSK when it was discovered that the drug was causing cancer. As well as adding Cardarine to its prohibited list of performance-enhancing substances in 2009, the World Anti-Doping Agency (WADA) also issued warnings to athletes that Cardarine is not safe. 

As we have said in all our previous articles on SARMs, you must be in no doubt about the potential risks and side effects of using these drugs, which much be balanced against their beneficial effects, which are real, when they are used properly and in the right contexts. Because the spectre of cancer has been raised here, that caveat applies with as much force, if not more.

We will note that there are serious flaws in the studies that showed Cardarine causes cancer. While the long-term effects of human use remain to be investigated, short-term clinical studies of Cardarine use have recorded no adverse side effects.

But no studies on rats confirmed the presence of tumors — even run at massive doses (30mg/kg) — when running GW0742.
